چکیده

Recent advances in our understanding of blood cell development have included the identification of a hematopoietic progenitor derived from endothelial cells and the possibility that the embryonic and fetal environment can reprogram gene expression in adult hematopoietic stem cells.

Effects of Recombinant Human Erythropoietin (rHuEpo) on the Blood Cells and Hematopoiesis in Spleen of Rats

Recombinant human erythropoietin is a glycoprotein hormone that stimulates erythropoiesis. In clonogenic assays of hematopoietic progenitors, high concen-trations of erythropoietin (Epo) increase CFU-E and diminish the number of granulocytes formed per culture plate. Fetal progenitors are more sensitive to these effects of Epo than progenitors from adults. We administered doses 50, 100, 200 IU/...

Bone Marrow Endosteal Mesenchymal Progenitors Depend on HIF Factors for Maintenance and Regulation of Hematopoiesis

Maintenance and differentiation of hematopoietic stem cells (HSCs) is regulated through cell-autonomous and non-cell-autonomous mechanisms within specialized bone marrow microenvironments. Recent evidence demonstrates that signaling by HIF-1α contributes to cell-autonomous regulation of HSC maintenance. By investigating the role of HIF factors in bone marrow mesenchymal progenitors, we found th...

The developmental program of human dendritic cells is operated independently of conventional myeloid and lymphoid pathways.

Two distinct dendritic cell (DC) subsets, conventional DCs (cDCs) and plasmacytoid DCs (pDCs), have been shown to develop via either the myeloid or the lymphoid pathway in murine hematopoiesis. Lineage-specific phenotypes or functions of "myeloid" and "lymphoid" DCs, however, still remain elusive. Furthermore, such analysis has been particularly difficult in humans, due to lack of an assay syst...

Transcriptional accessibility for genes of multiple tissues and hematopoietic lineages is hierarchically controlled during early hematopoiesis.

Hematopoietic stem cells (HSCs) maintain hematopoiesis by giving rise to all types of blood cells. Recent reports suggest that HSCs also possess the potential to generate nonhematopoietic tissues. To evaluate the underlying mechanisms in the commitment of HSCs into multitissue and multihematopoietic lineages, we performed oligonucleotide array analyses targeting for prospectively purified HSCs,...

Extramedullary Myelopoiesis in Malaria Depends on Mobilization of Myeloid-Restricted Progenitors by IFN-γ Induced Chemokines

Resolution of a variety of acute bacterial and parasitic infections critically relies on the stimulation of myelopoiesis leading in cases to extramedullary hematopoiesis. Here, we report the isolation of the earliest myeloid-restricted progenitors in acute infection with the rodent malaria parasite, Plasmodium chabaudi. The rapid disappearance of these infection-induced myeloid progenitors from...
